About Prelone
Overview
Prelone contains prednisolone, a glucocorticoid medicine. It is used for severe allergic conditions that have not responded to conventional treatment, as well as certain skin, eye, lung and rheumatic disorders. Other uses include certain adrenal, blood and kidney disorders, acute flare-ups of multiple sclerosis and solid organ rejection.
What it treats
Severe allergic conditions that have not responded to conventional treatment, including allergic rhinitis, asthma and allergic skin conditions. Certain severe skin diseases, including pemphigus and Stevens–Johnson syndrome. Certain forms of nephrotic syndrome, to reduce fluid retention or protein loss in urine. Adrenal insufficiency, congenital adrenal hyperplasia, cancer-related high blood calcium and nonsuppurative thyroiditis. Ulcerative colitis and regional enteritis during critical periods. Certain blood disorders, including autoimmune hemolytic anemia and selected platelet disorders. Acute leukemia and aggressive lymphomas. Acute flare-ups of multiple sclerosis. Certain inflammatory eye conditions and temporal arteritis. Selected respiratory conditions, including sarcoidosis, asthma and acute COPD flare-ups. Short-term additional treatment for arthritis and related inflammatory conditions; treatment of lupus and certain other rheumatic diseases. Certain tuberculosis complications alongside appropriate tuberculosis treatment; trichinosis affecting the nervous system or heart; and acute or chronic solid organ rejection.
Safety information
Side effects
Reported side effects include increased appetite, weight gain, nausea, abdominal bloating, headache, dizziness, sweating, facial redness and feeling unwell. Skin may become thin, bruise easily or heal more slowly. Menstrual changes and increased hair growth can occur. The label does not state how frequently these effects occur. Contact a doctor if you develop muscle weakness, swelling, hives, or changes in mood or mental health. Serious side effects include high blood pressure, fluid retention, potassium loss and heart failure in susceptible people. Prednisolone can raise blood sugar, increase the need for diabetes medicines, suppress adrenal gland function and slow growth in children. Other serious effects include stomach or intestinal ulcers that may bleed or perforate, inflammation of the pancreas, seizures, and increased pressure around the brain, usually after stopping treatment. Eye effects include cataracts, glaucoma and increased pressure within the eye. Bone and muscle effects include osteoporosis, fractures, bone damage, muscle loss and tendon rupture. Thickening of the heart muscle has been reported in premature infants. These effects warrant medical attention.
When to call your doctor
If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
Do not take this medicine if you have a systemic fungal infection or are allergic to prednisolone or any of its ingredients.
Interactions
Prednisolone can interact with the following medicines. Discuss them with your doctor: - Barbiturates, phenytoin, ephedrine and rifampin may speed up the breakdown of prednisolone and reduce its effect. - Cyclosporin and prednisolone may increase each other’s effects. Seizures have been reported with this combination. - Estrogens and ketoconazole may slow corticosteroid breakdown, increasing their effects or the risk of side effects. - Warfarin’s blood-thinning effect may change. Frequent blood-clotting tests may be needed. - Aspirin and other nonsteroidal anti-inflammatory drugs (NSAIDs) increase the risk of stomach and intestinal side effects. Aspirin requires particular caution in people with low prothrombin, a blood-clotting protein. Corticosteroids may also increase the removal of salicylates from the body. - Potassium-depleting medicines, including certain diuretics and amphotericin B, increase the risk of low potassium. Low potassium can increase the risk of abnormal heart rhythms in people taking digitalis glycosides. - Anticholinesterase medicines may cause severe weakness when combined with corticosteroids in people with myasthenia gravis. Your doctor may need to change treatment before starting prednisolone. - Diabetes medicines may need adjustment because corticosteroids can raise blood sugar. Tell your doctor before receiving vaccines or toxoids. Prolonged corticosteroid treatment may reduce the immune response to them, and live vaccines may pose additional risks because corticosteroids can increase the multiplication of some vaccine organisms. Routine vaccination may need to be postponed. Prednisolone may also suppress reactions to skin tests.

Storage
Store in the original carton to protect it from light. Check the pack for whether it must be refrigerated before first use. Keep all medicines out of the sight and reach of children.
