About Rezurock

Overview

Rezurock contains belumosudil, a kinase inhibitor, and comes as an oral tablet. It treats chronic graft-versus-host disease (chronic GVHD) in adults and children aged 12 years and older after at least two previous systemic treatments have failed. It blocks enzymes called ROCK kinases and has been shown in laboratory studies to reduce inflammatory responses and abnormal signals linked to tissue scarring.

Side effects

Common side effects include infections, weakness or lack of energy, nausea, diarrhea, shortness of breath, cough, swelling, bleeding, abdominal pain, muscle and joint pain, headache and high blood pressure. Common laboratory changes include decreased phosphate, decreased lymphocyte counts and increased gamma-glutamyl transferase. Contact your doctor if you develop an infection, bleeding or shortness of breath. Rezurock can harm an unborn baby. If you could become pregnant, use effective contraception and contact your doctor if you become pregnant during treatment.

Before taking this medicine

The label lists no formal contraindications. However, Rezurock should be avoided in people with moderate or severe liver impairment. Rezurock can harm an unborn baby. Women who could become pregnant should be advised of this risk and use effective contraception. Do not breastfeed while taking Rezurock.

Interactions

Tell your doctor about all medicines you take. Strong CYP3A inducers and proton pump inhibitors can require your prescriber to adjust your Rezurock treatment. Rezurock can also interact with medicines handled by proteins or enzymes called BCRP, OATP1B1, CYP1A2, CYP3A, P-gp or UGT1A1. Your doctor can check whether your medicines are affected. Some combinations should be avoided, particularly when small changes in medicine levels could cause serious harm. Others may require closer monitoring for side effects or adjustment of the other medicine by your prescriber.

Storage
