SUICIDALITY AND ANTIDEPRESSANT DRUGS
Antidepressants increased the risk of suicidal thoughts and behavior in children, adolescents, and young adults in short-term trials. These trials did not show an increase in the risk of suicidal thoughts and behavior with antidepressant use in subjects aged 65 and older.
In patients of all ages who are started on antidepressant therapy, monitor closely for worsening, and for emergence of suicidal thoughts and behaviors. Advise families and caregivers of the need for close observation and communication with the prescriber.

About Wellbutrin XL, Zyban
Overview
Wellbutrin XL and Zyban are prescription medicines containing bupropion. Wellbutrin XL is used to treat depression, while Zyban helps people stop smoking alongside support to quit. Bupropion is thought to work by increasing the activity of certain brain chemicals involved in mood and nicotine cravings.
What it treats
Major depressive disorder (Wellbutrin XL). Prevention of seasonal major depressive episodes in people with seasonal affective disorder (Wellbutrin XL). Smoking cessation, alongside behavioural support (Zyban).
Safety information
Side effects
Common side effects include dry mouth, nausea, constipation, headache, dizziness, trouble sleeping, anxiety, sweating and trembling. Serious side effects include seizures, increased blood pressure, new or worsening depression, suicidal thoughts, unusual changes in mood or behaviour, and severe allergic reactions. Contact a doctor promptly for marked agitation, hallucinations, or unusually elevated mood, increased activity and reduced need for sleep. Seek immediate medical help for a seizure, thoughts of suicide or self-harm, difficulty breathing, swelling of the face or throat, or a severe rash with blistering or peeling. Sudden eye pain, eye redness or changes in vision also need urgent medical assessment.

Before taking this medicine
Do not take Wellbutrin XL or Zyban if you are allergic to bupropion or any of the ingredients, have a seizure disorder, or have ever had anorexia nervosa or bulimia. Do not take them while abruptly stopping heavy alcohol use, benzodiazepines, barbiturates or anti-seizure medicines, as this increases the risk of seizures. Do not take Wellbutrin XL and Zyban together, or combine either with another medicine containing bupropion. Do not use bupropion with monoamine oxidase inhibitors (MAOIs), or within 14 days of stopping an MAOI. Certain medicines, including linezolid and intravenous methylene blue, also have MAOI effects and must not be combined with bupropion without specific medical management. A prescriber should assess suitability carefully if you have a significant head injury, a brain tumour, a history of stroke, liver or kidney problems, high blood pressure, heart disease, diabetes treated with medicines, bipolar disorder, or a risk of angle-closure glaucoma. Heavy alcohol use and other medicines that increase seizure risk also require review. Discuss use during pregnancy or breastfeeding with the prescriber. Monitor for worsening depression, suicidal thoughts or unusual changes in mood or behaviour, particularly when treatment starts or changes. Seek urgent medical help if suicidal thoughts develop.
Interactions
Wellbutrin XL and Zyban both contain bupropion and must not be taken together or with other bupropion-containing medicines. Bupropion must not be combined with monoamine oxidase inhibitors (MAOIs). This includes certain antidepressants, the antibiotic linezolid and intravenous methylene blue. A waiting period is required when switching between bupropion and an MAOI antidepressant; your doctor must manage this. Tell your doctor about: • Medicines that increase seizure risk, including some antidepressants, antipsychotics, tramadol, theophylline and oral or injected corticosteroids. • Medicines whose effects bupropion may alter, including certain antidepressants, antipsychotics, metoprolol and medicines for irregular heart rhythms. Bupropion may also reduce tamoxifen’s effectiveness. • Carbamazepine, phenytoin, phenobarbital, ritonavir, efavirenz or clopidogrel, which can change bupropion levels. • Levodopa or amantadine, which may increase nervous-system side effects. • Nicotine replacement products, which may increase the risk of high blood pressure when combined with bupropion. Minimise or avoid alcohol. Suddenly stopping heavy alcohol use, benzodiazepines, sedatives or antiseizure medicines can increase seizure risk. Discuss any planned changes with your doctor. Check with your doctor or pharmacist before starting any non-prescription medicine, herbal remedy or supplement.

Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
