About Saphnelo
Overview
Saphnelo (anifrolumab-fnia) is a monoclonal antibody medicine supplied in a vial for intravenous infusion. It treats adults with moderate to severe systemic lupus erythematosus (SLE) who are receiving standard therapy. It blocks signals from type I interferons, proteins involved in the immune and inflammatory processes that contribute to lupus. Its use is not recommended in people with severe active lupus affecting the kidneys or central nervous system, as its effectiveness has not been evaluated in these groups.
What it treats
• Moderate to severe systemic lupus erythematosus (SLE) in adults receiving standard therapy. Not recommended for severe active lupus nephritis or severe active central nervous system lupus, as effectiveness has not been evaluated in these situations.
Safety information
Side effects
Common side effects include nose and throat infections, upper respiratory tract infections, bronchitis, reactions related to the infusion, shingles and cough. Contact a doctor promptly if you develop a new infection during treatment. Serious and sometimes fatal infections have occurred with Saphnelo, and it increases the risk of respiratory infections and shingles. Seek immediate medical help for a serious allergic reaction, including anaphylaxis or angioedema (swelling under the skin). The label also includes a warning about cancer (malignancy). Discuss any known cancer risk factors with your doctor.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
Do not use Saphnelo if you have previously had anaphylaxis, a severe allergic reaction, to anifrolumab-fnia. Treatment should not start during an active infection. Serious and sometimes fatal infections have occurred with Saphnelo, and it increases the risk of respiratory infections and shingles (herpes zoster). Your prescriber should weigh the benefits and risks if you have severe or chronic infections and may interrupt treatment if a new infection develops. Serious allergic reactions, including anaphylaxis and angioedema (swelling beneath the skin), have been reported. Your prescriber should also weigh the benefits and risks if you have known risk factors for cancer. Avoid live or live-attenuated vaccines during treatment. Using Saphnelo with other biologic therapies is not recommended.
Interactions
No formal drug interaction trials have been conducted with Saphnelo.
