The use of benzodiazepines, including ONFI, exposes users to risks of abuse, misuse, and addiction, which can lead to overdose or death. Abuse and misuse of benzodiazepines commonly involve concomitant use of other medications, alcohol, and/or illicit substances, which is associated with an increased frequency of serious adverse outcomes. Before prescribing ONFI and throughout treatment, assess each patient’s risk for abuse, misuse, and addiction
The continued use of benzodiazepines, including ONFI, may lead to clinically significant physical dependence. The risks of dependence and withdrawal increase with longer treatment duration and higher daily dose. Abrupt discontinuation or rapid dosage reduction of ONFI after continued use may precipitate acute withdrawal reactions, which can be life-threatening. To reduce the risk of withdrawal reactions, use a gradual taper to discontinue ONFI or reduce the dosage

About Onfi
Overview
Onfi tablets contain clobazam, a benzodiazepine medicine. It is used alongside other treatments for seizures associated with Lennox-Gastaut syndrome in patients aged 2 years and older. Its exact action is not fully understood, but it is thought to strengthen the effects of GABA, a chemical messenger that reduces nerve activity in the brain.
What it treats
Seizures associated with Lennox-Gastaut syndrome (LGS) in patients aged 2 years or older, used alongside other seizure treatments.
Safety information
Side effects
Common side effects of Onfi (clobazam) include constipation, sleepiness or sedation, fever, low energy and drooling. Sedation may be increased when Onfi is taken with other medicines that slow brain and nervous system activity. Serious side effects include severe skin reactions, such as Stevens–Johnson syndrome and toxic epidermal necrolysis, and DRESS, a serious hypersensitivity reaction that can affect multiple organs. Stop taking Onfi and contact a doctor immediately at the first sign of a rash unless it is clearly unrelated to the medicine. Seek immediate medical help for suicidal thoughts or behaviour. Other serious risks include abuse, misuse, addiction, dependence and withdrawal reactions. Taking Onfi with opioids carries additional serious risks. Contact a doctor about severe sleepiness or concerns about dependence or withdrawal. Use during pregnancy can cause sedation or withdrawal symptoms in the newborn.

Before taking this medicine
Do not take Onfi (clobazam) if you have ever had an allergic reaction to clobazam or any of its ingredients. Onfi can cause sleepiness or sedation. This risk may increase when it is taken with other medicines that slow central nervous system activity. Serious skin reactions, including Stevens–Johnson syndrome and toxic epidermal necrolysis, can occur. Stop taking Onfi at the first sign of a rash unless it is clearly unrelated to the medicine, and seek medical advice promptly. A serious allergic reaction called DRESS, which can affect multiple organs, also requires stopping Onfi if no other cause is found. Monitor for suicidal thoughts or behaviour and seek medical help if these occur. If you are pregnant, discuss the risks with your prescriber. Animal studies suggest Onfi may harm the unborn baby. Use during pregnancy can cause sedation or withdrawal symptoms in the newborn.
Interactions
Tell your doctor if you drink alcohol, as it can increase clobazam levels in the blood. Also review your medicines with your doctor, especially those processed by the enzyme CYP2D6 or those that strongly or moderately block the enzyme CYP2C19. Your doctor may need to adjust Onfi or the other medicine.

Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
