About Gamifant
Overview
Gamifant (emapalumab-lzsg) is a monoclonal antibody medicine used to treat adults and children with certain forms of hemophagocytic lymphohistiocytosis (HLH). It treats primary HLH that has not responded to conventional treatment, has returned or is worsening, or when conventional treatment cannot be tolerated. It also treats HLH/macrophage activation syndrome (MAS) in known or suspected Still’s disease, including systemic juvenile idiopathic arthritis, when glucocorticoids have not worked well enough or cannot be tolerated, or when MAS returns. It works by binding to and blocking interferon gamma, a protein that may play a key role in HLH when produced in excessive amounts.
What it treats
- Primary hemophagocytic lymphohistiocytosis (HLH) in adults and children when the disease has not responded to treatment, has returned or is worsening, or when conventional HLH treatment cannot be tolerated. - HLH/macrophage activation syndrome (MAS) in adults and children with known or suspected Still’s disease, including systemic juvenile idiopathic arthritis (sJIA), when glucocorticoids have not worked adequately or cannot be tolerated, or when MAS has returned.
Safety information
Side effects
Common side effects in patients with primary HLH include infections, high blood pressure, infusion-related reactions and fever. In patients with HLH/MAS in Still’s disease, common side effects include viral infections, including cytomegalovirus (CMV) infection or reactivation, and rash. Infections and infusion-related reactions can be serious. Contact your doctor promptly if you develop signs or symptoms of an infection. Tell your healthcare team immediately if you experience a reaction during an infusion; the infusion may need to be interrupted and medical treatment provided. Do not receive live or live attenuated vaccines during treatment because of the increased risk of infection.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
The FDA label lists no contraindications for Gamifant. Infections require careful monitoring and prompt treatment. Patients should be tested for latent tuberculosis. Preventive treatment against herpes zoster, Pneumocystis jirovecii and fungal infections should be considered. Do not receive live or live attenuated vaccines while receiving Gamifant. Patients should be monitored for infusion-related reactions. If a reaction occurs, the infusion should be interrupted and appropriate medical care provided.

Storage
Store in the original carton to protect it from light. Check the pack for whether it must be refrigerated before first use. Keep all medicines out of the sight and reach of children.
