About Zelboraf
Overview
Zelboraf (vemurafenib) is a kinase inhibitor available as tablets. It treats melanoma that cannot be removed by surgery or has spread to other parts of the body, when an FDA-approved test detects a BRAF V600E mutation. It also treats Erdheim-Chester disease with a BRAF V600 mutation, but is not indicated for melanoma with non-mutated (wild-type) BRAF. It works by blocking certain mutated BRAF proteins that can cause cells to multiply.
What it treats
- Melanoma that cannot be removed by surgery or has spread to other parts of the body, with a BRAF V600E mutation confirmed by an FDA-approved test. Not indicated for melanoma without a BRAF mutation (wild-type BRAF). - Erdheim-Chester disease with a BRAF V600 mutation.
Safety information
Side effects
Common side effects include joint pain, rash, hair loss, tiredness and skin growths called papillomas. In people treated for melanoma, nausea, itching and increased sensitivity to sunlight are also common. In people treated for Erdheim-Chester disease, a prolonged QT interval, an electrical change in the heart that can affect its rhythm, is also common. Contact a doctor promptly about serious side effects, including new skin cancers or other cancers, liver injury, kidney failure, eye inflammation, or thickening and tightening of tissue in the hands or soles of the feet. Severe reactions associated with radiation treatment can also occur. Serious allergic reactions, including anaphylaxis and DRESS (a reaction that can affect several organs), and severe skin reactions such as Stevens-Johnson syndrome and toxic epidermal necrolysis require immediate medical attention. Heart rhythm changes, liver injury and kidney problems require monitoring with ECGs and blood tests. Regular skin checks are also needed. Avoid sun exposure because Zelboraf can increase sensitivity to sunlight. Zelboraf can harm an unborn baby; effective contraception is advised during treatment.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Interactions
Avoid taking Zelboraf with medicines that strongly block or increase the activity of the liver enzyme CYP3A4. Zelboraf can also increase blood levels of medicines broken down by the enzyme CYP1A2. Avoid combining it with these medicines when even a small increase in their levels could cause harmful effects. If the combination cannot be avoided, your doctor should monitor closely for side effects and may reduce the dose of the other medicine.

Storage
