About VYLOY
Overview
VYLOY (zolbetuximab-clzb) is an antibody medicine given into a vein. It is used with fluoropyrimidine- and platinum-containing chemotherapy as the first treatment for adults with stomach or gastroesophageal junction adenocarcinoma that has spread nearby and cannot be removed by surgery, or has spread to other parts of the body. The cancer must be HER2-negative and test positive for claudin 18.2 using an FDA-approved test. VYLOY targets claudin 18.2 on cells and activates immune processes that destroy those cells.
What it treats
First-line treatment of adults with HER2-negative, claudin 18.2-positive adenocarcinoma of the stomach or gastroesophageal junction that is locally advanced and cannot be removed by surgery, or has spread to other parts of the body. Used with fluoropyrimidine- and platinum-containing chemotherapy. Claudin 18.2 status must be confirmed by an FDA-approved test.
Safety information
Side effects
Common side effects reported when VYLOY was used with mFOLFOX6 or CAPOX chemotherapy include nausea, vomiting, tiredness, reduced appetite, diarrhea, numbness or tingling from nerve damage, abdominal pain, constipation, weight loss, allergic reactions and fever. Common blood test changes include reduced blood cell counts, low albumin, increased creatinine, raised liver enzymes, high or low blood sugar, and low sodium, phosphate, potassium or magnesium levels. Serious side effects include severe allergic reactions, including anaphylaxis, and reactions related to the infusion. Infusion-related reactions can be fatal. Tell your healthcare team immediately if you develop a reaction during or after an infusion. Patients should be monitored during the infusion and for at least two hours afterward. Severe nausea and vomiting also require prompt contact with your healthcare team. Medicines to prevent nausea and vomiting are given before each infusion. Severe reactions may require the infusion to be interrupted or treatment to be permanently stopped.

Before taking this medicine
The label lists no contraindications for VYLOY. Serious allergic reactions, including anaphylaxis, and serious or fatal infusion-related reactions have occurred. Patients must be monitored during the infusion and for at least 2 hours afterward. Depending on the reaction, the healthcare team may pause or slow the infusion, or permanently stop treatment. After an allergic reaction, antihistamines should be given before subsequent infusions. VYLOY can cause severe nausea and vomiting. Medicines to prevent nausea and vomiting should be given before each infusion. Additional treatment or fluid replacement may be needed during and after the infusion. Severe symptoms may require treatment to be paused or permanently stopped. Do not breastfeed during treatment.

Storage
