About Votrient

Overview

Votrient contains pazopanib, a prescription medicine in a group called kinase inhibitors. It is used to treat adults with advanced renal cell carcinoma, a type of kidney cancer, and advanced soft tissue sarcoma after prior chemotherapy. Its effectiveness has not been demonstrated for adipocytic soft tissue sarcoma or gastrointestinal stromal tumors. It works by blocking several proteins involved in cell signalling, including signals that help new blood vessels form.

What it treats

- Advanced renal cell carcinoma (kidney cancer) in adults. - Advanced soft tissue sarcoma in adults who have previously received chemotherapy. Effectiveness has not been demonstrated for adipocytic soft tissue sarcoma or gastrointestinal stromal tumors.

Side effects

Common side effects of pazopanib include diarrhoea, high blood pressure, loss of hair colour, nausea, reduced appetite and vomiting. In patients with soft tissue sarcoma, common side effects also include tiredness, weight loss, tumour pain, muscle and joint pain, headache, changes in taste, shortness of breath and lighter skin colour. Serious side effects warrant immediate contact with a doctor. These include severe liver damage; abnormal heart rhythms; reduced heart function or heart failure; severe bleeding; blood clots in arteries or veins, including clots in the lungs; and damage to small blood vessels called thrombotic microangiopathy. Other serious risks include a hole or abnormal connection in the stomach or intestines, lung inflammation, and a brain condition called posterior reversible encephalopathy syndrome (PRES). Several of these complications can be fatal. Pazopanib can also cause dangerously high blood pressure, an underactive thyroid, protein in the urine or nephrotic syndrome, impaired wound healing, and serious infections. Tumour lysis syndrome, a complication involving the breakdown of tumour cells, has been reported and can be fatal. Monitoring includes liver and thyroid tests, blood pressure checks and urine tests; heart monitoring may also be needed in patients at risk. Pazopanib can harm an unborn baby. Effective contraception is advised, and pregnancy during treatment should be reported to a doctor. Tell your doctor about planned surgery because treatment needs to be interrupted around surgery to reduce wound-healing risks.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.

Interactions

Some medicines can interact with Votrient (pazopanib). Discuss the following with your doctor: • Strong CYP3A4 inhibitors or inducers, medicines that affect a liver enzyme involved in processing drugs. Strong inhibitors should be avoided. An alternative to a strong inducer should be considered; pazopanib is not recommended if long-term use of a strong inducer cannot be avoided. • Medicines processed by CYP3A4, CYP2D6 or CYP2C8 that have only a small margin between effective and harmful levels. Combining these with pazopanib is not recommended. • Simvastatin. Taking it with pazopanib increases the risk of raised ALT, a liver enzyme. Weekly liver-function monitoring is recommended. Depending on the severity of any liver toxicity, your doctor may need to interrupt, adjust or stop pazopanib treatment. • Medicines that reduce stomach acid, including proton pump inhibitors and H2-receptor blockers. These should be avoided with pazopanib. Your doctor may consider a short-acting antacid instead and advise how to separate it from pazopanib.

Storage

Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
