About Verzenio

Overview

Verzenio (abemaciclib) is a tablet medicine used to treat adults with hormone receptor (HR)-positive, HER2-negative breast cancer. It is used with tamoxifen or an aromatase inhibitor for early breast cancer that involves lymph nodes and has a high risk of returning. For advanced or metastatic breast cancer, it is used with an aromatase inhibitor as initial hormone-based treatment, with fulvestrant after the cancer has progressed following hormone therapy, or alone after progression following hormone therapy and prior chemotherapy for metastatic disease. It works by blocking proteins called CDK4 and CDK6, which help breast cancer cells grow and divide.

What it treats

- HR-positive, HER2-negative early breast cancer in adults that has spread to lymph nodes and has a high risk of recurrence, as adjuvant treatment with tamoxifen or an aromatase inhibitor. - HR-positive, HER2-negative advanced or metastatic breast cancer in adults, used: - With an aromatase inhibitor as initial endocrine-based treatment. - With fulvestrant when the cancer has progressed after endocrine therapy. - Alone when the cancer has progressed after endocrine therapy and prior chemotherapy for metastatic disease.

Safety information

Side effects

Common side effects include diarrhea, nausea, abdominal pain, infections, tiredness, decreased appetite, vomiting, headache and hair loss. Low blood cell counts are also common, including low neutrophils and other white blood cells, low red blood cells (anemia) and low platelets. Serious side effects include severe diarrhea with dehydration and infection, low neutrophil counts, lung inflammation (which can be fatal), liver problems, and blood clots in the veins or lungs. Contact your doctor if you develop or suspect a serious side effect. At the first sign of loose stools, start antidiarrheal treatment, increase oral fluids and notify your healthcare provider. Blood counts and liver function need monitoring before and during treatment. Verzenio can harm an unborn baby. Use effective contraception and contact your doctor if you become pregnant.

When to call your doctor

- Contact your healthcare provider right away at the first sign of loose stools or diarrhea. Severe diarrhea can lead to dehydration and may occur with infection.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.

Before taking this medicine

The FDA label lists no contraindications for Verzenio (abemaciclib). However, it includes important warnings about severe diarrhea, low neutrophil counts (a type of white blood cell), lung inflammation, liver problems and blood clots, including clots in the lungs. At the first sign of loose stools, start antidiarrheal treatment, increase fluid intake and notify your healthcare provider. Diarrhea can be severe and may lead to dehydration and infection. Blood counts and liver function tests are needed before and regularly during treatment. Severe or fatal lung inflammation has been reported; severe cases require permanent discontinuation. Verzenio can harm an unborn baby. Use effective contraception during treatment and discuss any pregnancy with your healthcare provider. Do not breastfeed while taking Verzenio.

Interactions

Avoid taking Verzenio with ketoconazole or medicines classed as strong or moderate CYP3A inducers. Other strong or moderate CYP3A inhibitors require your prescriber to adjust your Verzenio treatment. Ask your doctor or pharmacist to check your medicines for these interactions before starting or stopping any medicine.

Storage
