About Vectibix
Overview
Vectibix contains panitumumab, a monoclonal antibody given into a vein to treat certain types of metastatic colorectal cancer in adults. For cancers without KRAS or NRAS mutations, it is used with FOLFOX chemotherapy as initial treatment, or alone after the cancer has progressed following specified chemotherapy treatments. For cancers with a KRAS G12C mutation confirmed by an FDA-approved test, it is used with sotorasib after prior chemotherapy. It works by blocking EGFR, a protein on cells that helps send signals for cell growth and survival.
What it treats
- Metastatic colorectal cancer in adults whose tumours have no mutations in either KRAS or NRAS (wild-type RAS), confirmed by an FDA-approved test. Used with FOLFOX chemotherapy as first-line treatment, or alone if the cancer has progressed after fluoropyrimidine-, oxaliplatin- and irinotecan-containing chemotherapy. - Metastatic colorectal cancer in adults whose tumours have a KRAS G12C mutation, confirmed by an FDA-approved test. Used with sotorasib after prior fluoropyrimidine-, oxaliplatin- and irinotecan-based chemotherapy. Not indicated when RAS mutation status is unknown, or for RAS-mutant metastatic colorectal cancer except with sotorasib for the KRAS G12C-mutated type.
Safety information
Side effects
Common side effects of Vectibix used alone include skin rash, inflammation around the nails, tiredness, nausea and diarrhea. When used with FOLFOX chemotherapy, common effects also include mouth sores, inflammation of moist body linings, weakness, loss of appetite, low magnesium or potassium levels, acne-like rash, itching and dry skin. With sotorasib, common effects include rash, dry skin, diarrhea, mouth sores, tiredness, and muscle or bone pain. Contact a doctor promptly for severe skin or soft tissue reactions, reactions during an infusion, or new or worsening eye problems. Serious eye problems include inflammation, ulceration or perforation of the cornea. Other serious risks include acute kidney failure, lung inflammation or scarring, and changes in blood mineral levels. Blood tests are needed to monitor these levels. Severe or life-threatening reactions may require treatment to be interrupted or stopped. Vectibix can increase sensitivity to sunlight; limit sun exposure. In patients whose metastatic colorectal cancer has RAS mutations, Vectibix used alone or with oxaliplatin-based chemotherapy may increase tumor progression or the risk of death, or provide no benefit. Combining Vectibix with bevacizumab and chemotherapy increases the risk of death and toxicity. Vectibix can harm an unborn baby. Use effective contraception during treatment and for 2 months after the last dose.
