About Tygacil

Overview

Tygacil contains tigecycline, an antibacterial medicine supplied in a vial for infusion into a vein. It is used in adults aged 18 and over to treat complicated skin and skin structure infections, complicated infections within the abdomen, and community-acquired bacterial pneumonia. Tigecycline acts against bacteria and belongs to the tetracycline class of antibiotics. Tygacil is not indicated for diabetic foot infections or hospital-acquired pneumonia, including ventilator-associated pneumonia.

What it treats

- Complicated infections of the skin and underlying tissues in adults aged 18 and over. - Complicated infections within the abdomen in adults aged 18 and over. - Community-acquired bacterial pneumonia in adults aged 18 and over.

Safety information

Side effects

Common side effects include nausea, vomiting, diarrhea, abdominal pain, headache and raised levels of the liver enzyme SGPT on blood tests. Serious risks that warrant contacting a doctor include life-threatening allergic reactions (anaphylaxis), liver problems, inflammation of the pancreas (pancreatitis), and low fibrinogen, a protein involved in blood clotting. Pancreatitis has been fatal in some cases. Contact a doctor if diarrhea occurs, as it may be associated with Clostridioides difficile. Blood-clotting tests, including fibrinogen, should be checked before treatment and regularly during treatment. Clinical trials found a higher risk of death from any cause in patients treated with Tygacil than in those receiving comparator treatments. The reason is not known. The greatest difference was seen in patients with ventilator-associated pneumonia. Higher mortality and lower cure rates were also reported in hospital-acquired pneumonia. Use during the last half of pregnancy, infancy or childhood up to age 8 may cause permanent tooth discoloration and underdeveloped tooth enamel. Use during the second or third trimester of pregnancy, infancy or childhood up to age 8 may also slow bone growth; this effect may be reversible.

When to call your doctor

- Seek immediate medical attention for a severe allergic reaction, which can be life-threatening. - Contact your doctor immediately if you develop abdominal pain. - Contact your doctor immediately if diarrhea occurs; it may need assessment for a Clostridioides difficile infection.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.

Before taking this medicine

Do not use Tygacil if you are allergic to tigecycline or any of its ingredients. It should also be avoided if you have a known allergy to tetracyclines. Severe, potentially life-threatening allergic reactions have been reported. Clinical trials found a higher risk of death in patients treated with Tygacil than in those receiving comparison treatments. The cause is unknown. The greatest difference was seen in patients with ventilator-associated pneumonia. Liver problems and pancreatitis, including fatal cases, have been reported. Abnormal liver tests require monitoring and reassessment of whether treatment should continue. If pancreatitis is suspected, the prescriber should consider stopping treatment. Tygacil can also lower fibrinogen, a protein involved in blood clotting. Blood clotting tests, including fibrinogen, should be checked before and regularly during treatment. Diarrhea during treatment requires assessment for Clostridioides difficile-associated diarrhea. Use during the last half of pregnancy, infancy or childhood up to age 8 may permanently discolor teeth and affect tooth enamel. Use during the second or third trimester of pregnancy, infancy or childhood up to age 8 may temporarily inhibit bone growth. Use in anyone under 18 is not recommended because of the higher risk of death seen in adult trials. Avoid breastfeeding for longer than 3 weeks while receiving Tygacil. An alternative is to pump and discard breast milk during treatment and for 9 days after the last dose.

Interactions

Tell your doctor if you take warfarin, tacrolimus or cyclosporine. If Tygacil is given with warfarin, blood tests to monitor clotting should be checked. Tygacil treatment also requires monitoring of tacrolimus or cyclosporine blood levels because of the risk of toxicity.
