About Truqap
Overview
Truqap (capivasertib) is a prescription cancer medicine for adults, supplied as tablets. It is used with fulvestrant for HR-positive, HER2-negative breast cancer that is locally advanced or has spread, has a PIK3CA, AKT1 or PTEN alteration confirmed by an FDA-authorized test, and has progressed after endocrine-based treatment for metastatic disease or returned during or within 12 months of completing adjuvant therapy. It is also used with abiraterone and prednisone for metastatic prostate cancer that is androgen pathway modulation-naïve or -sensitive and is PTEN-deficient, as confirmed by an FDA-authorized test. It works by blocking AKT proteins involved in signals that support cancer cell growth.
What it treats
- Hormone receptor (HR)-positive, HER2-negative breast cancer in adults that is locally advanced or has spread to other parts of the body, with one or more PIK3CA, AKT1 or PTEN alterations confirmed by an FDA-authorized test. Used with fulvestrant after progression on at least one endocrine-based treatment for metastatic disease, or recurrence during or within 12 months of completing adjuvant therapy. - PTEN-deficient prostate cancer in adults that has spread to other parts of the body and is androgen pathway modulation-naïve or -sensitive. PTEN deficiency must be confirmed by an FDA-authorized test. Used with abiraterone and prednisone.
Safety information
Side effects
Common side effects of Truqap include diarrhea, skin reactions and tiredness. Nausea, vomiting and mouth inflammation are also common in patients with breast cancer. Common blood test changes include increased blood sugar, triglycerides and creatinine, and reduced hemoglobin and certain white blood cells. In patients with prostate cancer, common changes also include low potassium, low sodium and increased liver enzymes. Truqap can cause severely high blood sugar, including diabetic ketoacidosis, which can be fatal. Blood sugar and HbA1c need checking before treatment and regularly during treatment. Contact your doctor if you develop high blood sugar or skin reactions. If diarrhea occurs, increase oral fluids, start antidiarrheal treatment and contact your healthcare provider. Depending on severity, these reactions may require your prescriber to interrupt, adjust or permanently stop treatment. Truqap can harm an unborn baby. Use effective contraception and contact your doctor if you become pregnant during treatment.

Before taking this medicine
Do not take Truqap if you have a severe allergy to capivasertib or any of its ingredients. Truqap can cause severely high blood sugar, including diabetic ketoacidosis, which can be fatal. Blood glucose and HbA1c should be checked before treatment and regularly during treatment. Diarrhea occurs in most patients. If it develops, increase fluid intake, start antidiarrheal treatment and contact your healthcare provider. Skin reactions also require monitoring. Depending on the severity of these side effects, your prescriber may interrupt, reduce or permanently stop treatment. Truqap can harm an unborn baby. Use effective contraception, and consult the fulvestrant prescribing information for its pregnancy and contraception guidance. Do not breastfeed during treatment.
Interactions
Tell your doctor about all medicines and supplements you take. TRUQAP should not be used with strong CYP3A inhibitors unless the combination cannot be avoided; in that case, your prescriber will need to reduce your TRUQAP dose. Moderate CYP3A inhibitors also require a dose reduction. Avoid using TRUQAP with strong or moderate CYP3A inducers. Do not change your dose yourself.

Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
