About Talzenna

Overview

Talzenna contains talazoparib, a PARP inhibitor supplied as capsules. It is used alone to treat adults with HER2-negative breast cancer that is locally advanced or has spread, when an FDA-approved test identifies a harmful or suspected harmful inherited BRCA mutation. It is also used with enzalutamide for adults with prostate cancer that has spread, has changes in HRR genes and no longer responds to treatment that lowers testosterone. It works by blocking PARP enzymes involved in DNA repair, which can lead to DNA damage and cancer cell death.

What it treats

- HER2-negative breast cancer in adults that is locally advanced or has spread to other parts of the body, with a harmful or suspected harmful inherited BRCA mutation confirmed by an FDA-approved companion diagnostic. Used on its own. - Prostate cancer in adults with an HRR gene mutation that has spread to other parts of the body and is resistant to treatment that lowers testosterone. Used with enzalutamide.

Side effects

Common side effects of Talzenna used alone include tiredness, nausea, headache, vomiting, hair loss, diarrhea and decreased appetite. Blood tests may show low hemoglobin, white blood cells or platelets, increased blood sugar and liver enzymes, and low calcium. When used with enzalutamide, common side effects include tiredness, nausea, decreased appetite, dizziness, changes in taste and bone fractures. Blood tests may show low hemoglobin, white blood cells or platelets, low calcium, sodium, phosphate, magnesium or potassium, and increased bilirubin. Serious side effects that warrant contacting a doctor include suppression of blood cell production, which can cause anemia, low white blood cells and low platelets. Myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML), serious bone marrow and blood disorders, have occurred in people taking Talzenna; some cases were fatal. Monitoring for blood-related toxicity is required, and treatment must be stopped if MDS or AML is confirmed. Talzenna can harm an unborn baby. Use effective contraception and contact your doctor if you become pregnant during treatment.

Before taking this medicine

The label lists no contraindications. Talzenna can cause fetal harm. Effective contraception is needed, and women should not breastfeed during treatment. Talzenna can reduce blood cell production, causing low red blood cell, white blood cell or platelet counts. Blood monitoring is required. Myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) have occurred, and some cases were fatal. Treatment must be stopped if MDS or AML is confirmed. Moderate or severe kidney impairment requires a prescriber-directed dose reduction and monitoring for increased side effects.

Interactions

Tell your doctor if you take medicines called P-gp inhibitors or BCRP inhibitors. These may increase side effects from Talzenna, so your doctor may monitor you more closely. Certain P-gp inhibitors also require your prescriber to reduce your Talzenna dose.

Storage

Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
