About Rybrevant
Overview
Rybrevant (amivantamab) is an antibody medicine given by intravenous infusion to treat adults with non-small cell lung cancer that is locally advanced or has spread to other parts of the body. It is used for cancers with specific EGFR gene changes, either alone or with other cancer medicines, depending on the gene change and previous treatment. It works by attaching to two proteins, EGFR and MET, disrupting their signals and helping immune cells target cancer cells for destruction.
What it treats
Rybrevant is used to treat adults with locally advanced or metastatic non-small cell lung cancer (NSCLC): • With EGFR exon 19 deletions or exon 21 L858R mutations, confirmed by an FDA-approved test: with lazertinib as first-line treatment. • With EGFR exon 19 deletions or exon 21 L858R mutations after the cancer has progressed during or after EGFR tyrosine kinase inhibitor treatment: with carboplatin and pemetrexed. • With EGFR exon 20 insertion mutations, confirmed by an FDA-approved test: with carboplatin and pemetrexed as first-line treatment. • With EGFR exon 20 insertion mutations, confirmed by an FDA-approved test, after the cancer has progressed during or after platinum-based chemotherapy: used alone.
Safety information
Side effects
Common side effects vary depending on whether Rybrevant is used alone or with lazertinib or carboplatin and pemetrexed. They include rash, nail problems or inflammation around the nails, infusion-related reactions, muscle and joint pain, tiredness, swelling, mouth sores, nausea, constipation and vomiting. Other common effects in some treatment groups include shortness of breath, cough, diarrhoea, reduced appetite, dry or itchy skin, tingling or numbness, bleeding, blood clots in veins and COVID-19. Blood tests may show serious changes, including low blood cell counts, low albumin, changes in mineral levels, raised liver enzymes or increased blood glucose. The changes reported differ depending on the treatment combination. Contact your doctor promptly about new or worsening breathing symptoms, which may indicate serious lung inflammation, or worsening eye symptoms, which need prompt assessment by an eye specialist. Severe skin reactions can occur, including toxic epidermal necrolysis, a severe skin condition; contact your doctor promptly about a severe or worsening rash. Tell the infusion team immediately about any reaction during an infusion. Blood clots in veins are an important risk when Rybrevant is used with lazertinib and require medical attention. Your doctor may recommend preventive blood-thinning treatment. Rybrevant can harm an unborn baby. Use effective contraception if you could become pregnant, and contact your doctor if you become pregnant during treatment.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
The FDA label lists no contraindications for Rybrevant. However, important precautions apply: • Infusion-related reactions: The infusion must be interrupted at the first sign of a reaction. Treatment may need to be slowed or permanently stopped, depending on severity. • Lung inflammation: New or worsening symptoms require monitoring. Treatment must be withheld if interstitial lung disease or pneumonitis is suspected and permanently stopped if confirmed. • Blood clots when used with lazertinib: Preventive anticoagulant treatment is recommended during the first four months. Blood clots may require treatment interruption or permanent discontinuation of Rybrevant. • Severe skin reactions: These can include acne-like inflammation and toxic epidermal necrolysis. Preventive medicines are recommended when treatment starts. Severe reactions may require treatment changes or discontinuation. • Eye problems: Worsening eye symptoms require prompt assessment by an eye specialist and may require treatment changes or discontinuation. • Pregnancy: Rybrevant can harm an unborn baby. Women who could become pregnant should use effective contraception. • Breastfeeding: Do not breastfeed during treatment.

Storage
Store in the original carton below 25°C, protected from light, unless the pack states refrigeration. Do not freeze. Keep all medicines out of the sight and reach of children.
