About Qinlock
Overview
Qinlock contains ripretinib, a medicine called a kinase inhibitor. It is used to treat adults with advanced gastrointestinal stromal tumor (GIST) who have previously received at least three kinase inhibitors, including imatinib. It works by blocking the activity of proteins called KIT and PDGFRA, including certain altered forms of these proteins.

Safety information
Side effects
Common side effects of Qinlock include hair loss, tiredness, nausea, abdominal pain, constipation, muscle pain, diarrhoea, reduced appetite, vomiting, and hand-foot syndrome, which affects the palms and soles. Blood tests may also show increased lipase or low phosphate levels, which can be severe. Contact your doctor if you develop hand-foot symptoms, new or changing skin lesions, or a reaction to sunlight. Serious risks include new skin cancers, high blood pressure, heart dysfunction and impaired wound healing. Skin checks, blood pressure monitoring and heart function assessments are needed before and during treatment, as directed by your doctor. Limit direct exposure to ultraviolet light and tell your doctor about any planned surgery. Qinlock can harm an unborn baby. If you could become pregnant, use effective contraception and contact your doctor if you become pregnant during treatment.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
The FDA label lists no formal contraindications for Qinlock. However, treatment should not be started if high blood pressure is uncontrolled. Qinlock requires caution because it can cause hand-foot skin reactions, new skin cancers, high blood pressure, heart dysfunction, impaired wound healing and sensitivity to sunlight. Skin examinations are needed before and regularly during treatment. Blood pressure should be monitored, and heart pumping function should be assessed before treatment and during treatment when clinically indicated. Severe heart dysfunction requires permanent discontinuation. Tell your healthcare team about planned surgery. Qinlock must be withheld around surgery and should not be restarted after major surgery until adequate wound healing. Limit direct exposure to ultraviolet light. Qinlock can harm an unborn baby. Women who could become pregnant should use effective contraception. Do not breastfeed during treatment.
Interactions
Strong CYP3A inhibitors can require more frequent monitoring for side effects while taking Qinlock. Strong or moderate CYP3A inducers should be avoided with Qinlock. If a moderate inducer cannot be avoided, your prescriber will need to adjust your treatment. Ask your doctor to check whether any medicines you take belong to these groups.

Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
