About Mektovi
Overview
Mektovi (binimetinib) is a prescription kinase inhibitor supplied as tablets. It is used with encorafenib to treat melanoma that cannot be removed by surgery or has spread and has a BRAF V600E or V600K mutation. The combination also treats adults with non-small cell lung cancer that has spread and has a BRAF V600E mutation; an FDA-approved test must confirm the relevant mutation for either use. Binimetinib blocks proteins called MEK1 and MEK2, which help carry signals involved in cancer cell growth.
What it treats
- Melanoma that cannot be removed by surgery or has spread, with a BRAF V600E or V600K mutation confirmed by an FDA-approved test. Used in combination with encorafenib. - Non-small cell lung cancer (NSCLC) that has spread in adults, with a BRAF V600E mutation confirmed by an FDA-approved test. Used in combination with encorafenib.
Safety information
Side effects
Common side effects of Mektovi used with encorafenib include tiredness, nausea, diarrhea, vomiting and abdominal pain. In patients treated for non-small cell lung cancer, common side effects also include muscle, bone or joint pain, vision problems, constipation, shortness of breath, rash and cough. Serious side effects that warrant prompt medical attention include weakened heart function, blood clots in the legs or lungs, eye problems affecting the retina or causing inflammation inside the eye, lung inflammation, liver injury, muscle breakdown and major bleeding. Contact your doctor promptly about any vision changes or new or worsening unexplained breathing symptoms. New cancers, including skin cancers, can occur with Mektovi and encorafenib. Monitoring for new cancers is needed before, during and after treatment. Heart function, eye examinations and blood tests are also needed to monitor for serious side effects. Mektovi can harm an unborn baby. Use effective contraception if you can become pregnant, and contact your doctor if you become pregnant during treatment.

Before taking this medicine
The FDA label lists no contraindications for Mektovi (binimetinib). However, it includes important warnings and monitoring requirements. Conditions requiring caution include reduced heart function, blood clots in the legs or lungs, eye problems, lung inflammation, liver injury, severe muscle breakdown and serious bleeding. Heart function should be checked before and during treatment; safety has not been established in patients whose left ventricular ejection fraction (LVEF), a measure of heart pumping function, is below 50%. Regular eye examinations and blood tests to monitor liver function, muscle injury and kidney function are also needed. New or worsening unexplained lung symptoms and any vision changes require assessment. When used with encorafenib, Mektovi can be associated with new cancers, including skin cancers. Monitoring is needed before, during and after treatment. Mektovi can harm an unborn baby. People who can become pregnant should use effective contraception. Do not breastfeed during treatment.
Interactions
