About LUMAKRAS
Overview
LUMAKRAS (sotorasib) is a medicine that targets cancers with a specific mutation called KRAS G12C, confirmed by an FDA-approved test. It is used alone for adults with locally advanced or metastatic non-small cell lung cancer after at least one prior systemic treatment; this use has accelerated approval, with continued approval dependent on confirmatory evidence of clinical benefit. It is also used with panitumumab for adults with metastatic colorectal cancer previously treated with fluoropyrimidine-, oxaliplatin- and irinotecan-based chemotherapy. It works by locking the KRAS G12C protein in an inactive state, blocking signals that help cancer cells grow.
What it treats
- KRAS G12C-mutated non-small cell lung cancer that is locally advanced or has spread, in adults who have received at least one prior systemic therapy. Used alone. This indication has accelerated approval; continued approval may depend on confirmatory trials. - KRAS G12C-mutated colorectal cancer that has spread, in adults previously treated with fluoropyrimidine-, oxaliplatin- and irinotecan-based chemotherapy. Used with panitumumab. For both uses, the KRAS G12C mutation must be confirmed by an FDA-approved test.
Safety information
Side effects
When LUMAKRAS is used alone for non-small cell lung cancer, common side effects include diarrhea, muscle and bone pain, nausea, tiredness, liver toxicity and cough. When used with panitumumab for colorectal cancer, common side effects include rash, dry skin, diarrhea, mouth inflammation, tiredness, and muscle and bone pain. Blood and urine tests may show changes in blood cell counts, liver enzymes, mineral levels or urine protein. Your doctor will monitor liver function during treatment. Serious side effects include liver injury and interstitial lung disease or pneumonitis (lung inflammation). Contact your doctor promptly if tests show liver problems or if you develop new or worsening breathing symptoms. These serious reactions may require treatment to be paused or permanently stopped by your doctor.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
The FDA label lists no contraindications for LUMAKRAS. LUMAKRAS can cause liver injury. Liver function tests should be monitored every 3 weeks for the first 3 months, then once monthly as clinically indicated. Depending on severity, the prescriber may interrupt, adjust or permanently stop treatment and consider systemic corticosteroids. Monitor for new or worsening breathing symptoms, which may indicate interstitial lung disease or pneumonitis. Treatment must be withheld immediately if either is suspected and permanently stopped if no other cause is identified. Do not breastfeed while taking LUMAKRAS.
Interactions
Tell your doctor about all medicines you take, particularly: • Acid-reducing medicines: Proton pump inhibitors (PPIs) and H2 receptor blockers should be avoided with LUMAKRAS. If an acid-reducing medicine cannot be avoided, ask your doctor how to separate LUMAKRAS from a local antacid. • Strong CYP3A4 inducers: These medicines should be avoided with LUMAKRAS. • Certain CYP3A4 substrates: LUMAKRAS may reduce their effectiveness. Combinations where even small changes in medicine levels could cause treatment failure should be avoided. • Certain P-gp substrates: LUMAKRAS may increase the risk of serious toxicity. Combinations where even small changes in medicine levels could cause serious harm should be avoided. If an affected CYP3A4 or P-gp substrate must be used, your doctor may need to adjust that medicine’s dose according to its prescribing information.

Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
