About Lorbrena
Overview
Lorbrena contains lorlatinib, a kinase inhibitor available as tablets. It is used to treat adults with non-small cell lung cancer that has spread to other parts of the body and is ALK-positive, as confirmed by an FDA-approved test. It works by blocking the activity of ALK, including some altered forms of this enzyme.
What it treats
Metastatic non-small cell lung cancer (NSCLC) in adults whose tumors are anaplastic lymphoma kinase (ALK)-positive, as detected by an FDA-approved test.
Safety information
Side effects
Common side effects include swelling, nerve problems such as numbness or tingling in the hands or feet, weight gain, changes in thinking, tiredness, shortness of breath, joint pain, diarrhea, mood changes, cough, and high cholesterol or triglyceride levels. Contact your doctor promptly about seizures; hallucinations or other psychotic symptoms; changes in thinking, speech, alertness or sleep; or mood changes, including thoughts of suicide. Seek immediate medical help for suicidal thoughts or seizures. Other serious risks include changes in the heart’s electrical conduction, lung inflammation, high blood pressure, high blood sugar, and serious liver injury when Lorbrena is taken with medicines that strongly increase CYP3A enzyme activity. Contact your doctor promptly if you develop new or worsening shortness of breath or cough. Blood pressure and blood sugar need monitoring during treatment. Lorbrena can harm an unborn baby. Contact your doctor if pregnancy occurs or is suspected during treatment.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
Do not take Lorbrena (lorlatinib) with strong CYP3A inducers, as this combination can cause serious liver damage. Your prescriber must arrange for these medicines to be stopped before Lorbrena is started. Lorbrena requires monitoring for seizures; changes in thinking, mood, speech, mental state or sleep; psychotic symptoms; and suicidal thoughts. Other important risks include increased blood fats, heart conduction problems (atrioventricular block), lung inflammation, high blood pressure and high blood sugar. Blood pressure and fasting blood sugar should be monitored during treatment, with fasting blood sugar also checked before treatment starts. These problems may require treatment to be interrupted, adjusted or stopped by your prescriber. Lorbrena can harm an unborn baby. Males and females who could conceive a child should use effective non-hormonal contraception. Do not breastfeed during treatment.
Interactions
Lorbrena must not be taken with strong CYP3A inducers. Use with moderate CYP3A inducers, strong CYP3A inhibitors or fluconazole should be avoided. If these combinations cannot be avoided, your prescriber may need to adjust your treatment. Also avoid combining Lorbrena with certain medicines processed by CYP3A or transported by P-gp when even small changes in their levels could cause serious treatment failure. Ask your doctor to check whether any of your medicines fall into these groups.

Dosing
Dosing is set by your prescriber. Each health care professional may prescribe medication differently according to a patient's medical history and condition, so follow the directions on your prescription label and speak to your doctor or pharmacist before changing anything.
Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
