Kyprolis

Before taking this medicine

The FDA label lists no contraindications for Kyprolis (carfilzomib). However, serious risks require careful monitoring and may require treatment to be paused or stopped. Conditions requiring caution include heart failure or reduced blood flow to the heart, kidney problems, liver impairment, high blood pressure, pulmonary hypertension, serious lung problems or severe breathlessness. Blood pressure, kidney function, liver enzymes and platelet counts should be monitored. Other serious risks include tumor lysis syndrome, blood clots, infusion-related reactions, severe or fatal bleeding, low platelet counts and damage to small blood vessels called thrombotic microangiopathy. New or worsening neurological or visual symptoms need prompt assessment because of the risks of posterior reversible encephalopathy syndrome (PRES) and progressive multifocal leukoencephalopathy (PML). Kyprolis can harm an unborn baby. Effective contraception is advised for females who could become pregnant and males with female partners who could become pregnant. Women should not breastfeed during treatment. Adverse reactions were more frequent in patients aged 75 years or older. Patients with liver impairment or receiving hemodialysis need treatment adjustments directed by their clinician. Combining Kyprolis with melphalan and prednisone in newly diagnosed patients who are not eligible for transplant increases the risk of serious and fatal toxicities.
