About Jemperli
Overview
Jemperli (dostarlimab) is an antibody medicine used with carboplatin and paclitaxel, then continued alone, to treat adults with endometrial cancer that is advanced at diagnosis or has returned. It is also used alone for recurrent or advanced endometrial cancer confirmed by an FDA-approved test to be mismatch repair deficient (dMMR), after progression during or after platinum-containing treatment, when curative surgery or radiation is not an option. Jemperli can also treat adults with other recurrent or advanced dMMR solid tumors confirmed by an FDA-approved test, after progression during or after prior treatment and when no satisfactory alternatives remain; this use has accelerated approval, with continued approval potentially dependent on confirmatory trials verifying clinical benefit. It works by blocking PD-1, helping release a brake on the immune system’s response against tumors.
What it treats
• Primary advanced or recurrent endometrial cancer in adults, used with carboplatin and paclitaxel, then continued alone. • Recurrent or advanced mismatch repair deficient (dMMR) endometrial cancer in adults, confirmed by an FDA-approved test, that has progressed during or after platinum-containing treatment, when curative surgery or radiation is not an option. Used alone. • Recurrent or advanced dMMR solid tumors in adults, confirmed by an FDA-approved test, that have progressed during or after prior treatment, with no satisfactory alternative treatment options. Used alone. This indication has accelerated approval based on tumor response and how long it lasts; continued approval may depend on confirmation of clinical benefit.
Safety information
Side effects
Common side effects of Jemperli (dostarlimab) used alone in patients with mismatch repair-deficient (dMMR) solid tumors include tiredness or weakness, anemia, diarrhea and nausea. When used with carboplatin and paclitaxel in patients with endometrial cancer, common side effects include tiredness, nausea, vomiting, hair loss, nerve problems in the hands or feet, joint pain, rash, constipation, diarrhea, abdominal pain, shortness of breath, reduced appetite and urinary tract infections. Common blood test changes include reduced blood cell counts, increased blood sugar, changes in liver and kidney tests, and reduced levels of certain minerals and albumin, a blood protein. With Jemperli used alone, severe laboratory abnormalities can include low lymphocyte counts, low sodium, increased alkaline phosphatase and low albumin. Contact your doctor promptly about suspected serious side effects. Jemperli can cause immune-related reactions in any organ or tissue. These may be severe or fatal and include inflammation of the lungs, bowel, liver or kidneys, hormone gland disorders, serious skin reactions and rejection of a transplanted organ. Blood tests to check liver, kidney and thyroid function are needed before and during treatment. Reactions can also occur during the infusion. Tell the treatment team immediately if you develop a reaction. Serious or fatal complications can occur in people who receive a donor stem cell transplant before or after treatment. Jemperli can harm an unborn baby. Use effective contraception if you could become pregnant, and contact your doctor if pregnancy occurs.

Before taking this medicine
The FDA label lists no contraindications for Jemperli (dostarlimab). However, important warnings apply: Jemperli can cause severe or fatal immune reactions affecting any organ or tissue, including the lungs, bowel, liver, hormone-producing glands, kidneys and skin. It can also cause rejection of a transplanted organ. Monitoring includes liver enzymes, kidney function and thyroid function before and during treatment. Depending on severity, treatment may need to be paused or permanently stopped, and corticosteroids may be needed. Infusion-related reactions can occur and may require the infusion to be slowed, interrupted or permanently stopped. Serious or fatal complications can occur in people who receive a stem cell transplant from a donor before or after treatment. Jemperli can harm an unborn baby. People who could become pregnant should use effective contraception. Do not breastfeed during treatment.
