About Jaypirca
Overview
Jaypirca contains pirtobrutinib, a kinase inhibitor supplied as tablets. It treats adults with mantle cell lymphoma that has returned or not responded to treatment after at least two prior systemic therapies, including a BTK inhibitor; this use has accelerated approval based on response rate, with continued approval potentially depending on confirmation of clinical benefit. It also treats adults with chronic lymphocytic leukemia or small lymphocytic lymphoma that has returned or not responded to treatment, following prior treatment with a covalent BTK inhibitor. It works by blocking BTK, a protein involved in signals that support B-cell growth and movement.
What it treats
- Mantle cell lymphoma (MCL) in adults whose disease has returned or has not responded after at least two lines of systemic treatment, including a BTK inhibitor. This use has accelerated approval based on response rate; continued approval may depend on a confirmatory trial verifying clinical benefit. - Chronic lymphocytic leukemia or small lymphocytic lymphoma (CLL/SLL) in adults whose disease has returned or has not responded to treatment and who have previously received a covalent BTK inhibitor.
Safety information
Side effects
Common side effects include tiredness and blood test changes: reduced white blood cells (including neutrophils and lymphocytes), platelets, hemoglobin and calcium. Serious side effects include infections, bleeding, low blood cell counts, abnormal heart rhythms such as atrial fibrillation or atrial flutter, and liver injury. Contact your doctor promptly if you develop signs of infection, bleeding or an abnormal heartbeat. Blood counts and liver function need monitoring during treatment. Other cancers, including skin cancers, have developed during treatment. Your doctor should monitor for these; use sun protection. Jaypirca can harm an unborn baby. If you could become pregnant, use effective contraception and contact your doctor if you become pregnant.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
The FDA label lists no contraindications for Jaypirca. However, treatment requires monitoring for infections, bleeding, low blood cell counts, abnormal heart rhythms, and liver injury. Blood counts and liver function should be checked during treatment. Other cancers, including skin cancers, have developed during treatment. Monitoring and sun protection are advised. Jaypirca can harm an unborn baby. Women who could become pregnant should use effective contraception. Do not breastfeed during treatment.
Interactions
Jaypirca can interact with medicines that strongly block the enzyme CYP3A or moderately or strongly increase its activity. These combinations should be avoided. If certain combinations cannot be avoided, your prescriber may need to adjust your treatment. Jaypirca can also affect certain medicines processed by CYP2C8, CYP2C19 or CYP3A, or transported by P-gp or BCRP. For some of these medicines, even small changes in blood levels may increase the risk of side effects. Ask your doctor to check your other medicines for interactions and follow the guidance in their prescribing information.

Storage
