About Inlyta
Overview
Inlyta (axitinib) is a prescription medicine called a kinase inhibitor, used to treat advanced renal cell carcinoma, a type of kidney cancer. It is used with avelumab or pembrolizumab as a first treatment, or on its own after one previous systemic treatment has failed. It works by blocking receptors involved in the formation of blood vessels that support tumour growth.

Side effects
Common side effects of Inlyta (axitinib) when taken alone include diarrhoea, high blood pressure, tiredness, weakness, reduced appetite, nausea, vomiting, constipation, weight loss, voice changes, and hand-foot syndrome, which affects the skin on the palms and soles. When Inlyta is taken with avelumab or pembrolizumab, common side effects also include an underactive thyroid, liver problems, rash, cough, and inflammation of the mouth or other moist body linings. With avelumab, muscle and bone pain, shortness of breath, abdominal pain, and headache are also common. Contact a doctor promptly about side effects, particularly serious problems such as: • Very high blood pressure, including a blood pressure crisis. • Blood clots in arteries or veins. • Bleeding. • Heart failure or other major heart and blood vessel problems, particularly with avelumab. • A hole in the stomach or intestine, or an abnormal connection between organs. • Liver injury. Severe liver enzyme increases occur more frequently when Inlyta is combined with avelumab or pembrolizumab. • Protein in the urine, thyroid problems, or poor wound healing. • Reversible posterior leukoencephalopathy syndrome (RPLS), a serious condition affecting the brain. Blood clots, bleeding, heart failure, and holes or abnormal connections in the digestive tract can be fatal. Blood pressure, thyroid function, urine protein, and liver tests need monitoring because some problems may be detected through checks rather than noticeable symptoms. Serious side effects may require treatment to be interrupted or stopped by the prescriber.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
The FDA label lists no formal contraindications. However, Inlyta should not be used in people with untreated brain metastases or recent active gastrointestinal bleeding. Blood pressure should be well controlled before treatment. Caution and monitoring are needed for people at increased risk of blood clots, bleeding, heart failure, or a hole or abnormal connection in the stomach or intestines. Thyroid function, urine protein and liver function should be checked before and during treatment. Inlyta can impair wound healing. Tell your prescriber about planned surgery or wounds that are not healing, as treatment may need to be interrupted. Moderate liver impairment requires a prescribing adjustment; Inlyta has not been studied in severe liver impairment. Inlyta can cause a serious neurological condition called reversible posterior leukoencephalopathy syndrome (RPLS), which requires permanent discontinuation. Combining Inlyta with avelumab or pembrolizumab can increase the risk of severe liver problems. The combination with avelumab also carries a warning for major cardiovascular events. Inlyta can harm an unborn baby. Use effective contraception during treatment. Do not breastfeed while taking Inlyta.
Interactions
Tell your doctor about all medicines and supplements you take. Inlyta should not be used with strong inhibitors or inducers of enzymes called CYP3A4/5. If a strong inhibitor cannot be avoided, your prescriber will need to adjust your Inlyta treatment.

Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
