About Imcivree
Overview
Imcivree (setmelanotide) is a prescription medicine supplied in a vial for injection under the skin. It is used to reduce excess body weight and maintain weight loss long term in adults and children aged 4 years and older with acquired hypothalamic obesity. It is also used for these purposes in adults and children aged 2 years and older with Bardet-Biedl syndrome or POMC, PCSK1 or LEPR deficiency confirmed by qualifying genetic test results. It works by activating MC4 receptors in the brain, which help regulate hunger, fullness and energy use.
What it treats
Imcivree (setmelanotide) is indicated to reduce excess body weight and maintain weight reduction long term in: • Adults and children aged 4 years and older with acquired hypothalamic obesity. • Adults and children aged 2 years and older with obesity due to Bardet-Biedl syndrome. • Adults and children aged 2 years and older with obesity due to POMC, PCSK1 or LEPR deficiency, confirmed by genetic testing showing variants classified as pathogenic, likely pathogenic or of uncertain significance.
Safety information
Side effects
Common side effects include skin darkening, injection site reactions, nausea, headache, diarrhea, abdominal pain, vomiting, depression and spontaneous penile erections. Serious side effects and changes that need medical attention include: - New or worsening depression, suicidal thoughts or suicidal behavior. Contact a doctor promptly. - Serious allergic reactions, including anaphylaxis. Stop Imcivree and seek medical attention immediately if a serious allergic reaction is suspected. - An erection lasting longer than 4 hours. Seek emergency medical attention. Changes in sexual arousal can also occur in females. - Darkening of existing moles or the development of new moles. Tell your doctor about these changes. A full-body skin examination is needed before treatment and periodically during treatment. - Acute adrenal insufficiency in patients with acquired hypothalamic obesity. Your doctor should monitor for signs of this condition. - Low or high blood sodium levels in patients with acquired hypothalamic obesity and central diabetes insipidus. Your doctor should monitor for signs and symptoms of these imbalances.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
Do not use Imcivree if you have previously had a serious allergic reaction to setmelanotide or any of its other ingredients. Serious reactions, including anaphylaxis, have been reported. If a serious allergic reaction is suspected, stop using Imcivree and seek medical attention promptly. Imcivree is not recommended while breastfeeding. Conditions and effects requiring caution include: - Depression or suicidal thoughts or behaviour: monitor for new or worsening symptoms and contact your prescriber if they occur. Treatment may need to be stopped. - Changes in sexual arousal: spontaneous erections and sexual side effects in females can occur. Seek emergency medical attention for an erection lasting longer than 4 hours. - Skin darkening or changes in moles: a full-body skin examination is needed before treatment and periodically during treatment. - Acquired hypothalamic obesity: monitoring is needed for signs of acute adrenal insufficiency. - Acquired hypothalamic obesity with central diabetes insipidus: monitoring is needed for signs and symptoms of low or high blood sodium.
