About Imbruvica

Overview

Imbruvica contains ibrutinib, a medicine called a kinase inhibitor. It treats adults with chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL), including those with a 17p deletion, and adults with Waldenström’s macroglobulinemia. It also treats chronic graft-versus-host disease in adults and children aged 1 year and older after one or more systemic treatments have failed. It works by blocking Bruton’s tyrosine kinase (BTK), a protein involved in signals that help B cells survive and move.

What it treats

- Chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL) in adults, including those with 17p deletion. - Waldenström’s macroglobulinemia (WM) in adults. - Chronic graft-versus-host disease (cGVHD) in adults and children aged 1 year and older after one or more systemic treatments have failed.

Safety information

Side effects

Common side effects in people with B-cell cancers include low platelet counts, diarrhea, tiredness, muscle or joint pain, low white blood cell counts, rash, anemia, bruising and nausea. In adults and children with chronic graft-versus-host disease (cGVHD), common side effects also include fever, muscle spasms, mouth inflammation, bleeding, abdominal pain, pneumonia and headache. Contact a doctor promptly about bleeding, fever or signs of infection, or symptoms of an abnormal heart rhythm or heart failure. Serious risks include severe bleeding, infections, abnormal heart rhythms, heart failure and sudden death. Other serious risks include high blood pressure, low blood cell counts, new cancers including skin cancer, liver injury and tumor lysis syndrome, a complication of rapid cancer cell breakdown. Blood pressure, blood cell counts and liver function need monitoring during treatment. Imbruvica can harm an unborn baby; effective contraception is needed for females who could become pregnant.

When to call your doctor

- Bleeding. - Fever or signs of infection. - An irregular heartbeat.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.

Before taking this medicine

The FDA label lists no formal contraindications for Imbruvica. However, use should be avoided in people with severe liver impairment. Mild or moderate liver impairment requires a dose adjustment by the prescriber. Imbruvica requires caution because of risks of bleeding, infections, abnormal heart rhythms, heart failure, sudden death, high blood pressure, low blood cell counts, liver injury and tumor lysis syndrome. Other cancers, including skin cancers, have also occurred. Monitoring includes blood pressure, monthly complete blood counts and liver function throughout treatment. Fever or signs of infection need prompt assessment. Imbruvica can harm an unborn baby. Women who could become pregnant should use effective contraception. Do not breastfeed during treatment.

Interactions

Tell your doctor if you take medicines called CYP3A inhibitors, as your prescriber may need to adjust your Imbruvica dose. Strong CYP3A inducers should not be taken with Imbruvica. Ask your doctor to check your medicines for these interactions.

Storage

Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
