About Glyxambi

Overview

Glyxambi is a prescription tablet containing empagliflozin and linagliptin. It is used alongside diet and exercise to improve blood sugar control in adults with type 2 diabetes. Its two ingredients help control blood sugar by blocking different targets in the body, called SGLT2 and DPP-4.

What it treats

- Improving blood sugar control in adults with type 2 diabetes, alongside diet and exercise. - The empagliflozin component is also indicated to reduce the risk of cardiovascular death in adults with type 2 diabetes and established cardiovascular disease.

Safety information

Side effects

Common side effects include urinary tract infections, inflammation of the nose and throat, and upper respiratory tract infections. Serious side effects and risks that need medical attention include: • Ketoacidosis, which can occur regardless of blood sugar level. • Acute pancreatitis (inflammation of the pancreas), including fatal cases. • Fluid depletion, particularly in older people, those with impaired kidney function or those taking loop diuretics. • Urinary and genital infections, including kidney infections, infections leading to sepsis, genital fungal infections and Fournier’s gangrene, a serious infection of tissue around the genitals and the area between the genitals and anus. Seek immediate medical attention for pain, tenderness, redness or swelling in this area with fever or feeling unwell. • Low blood sugar when used with insulin or medicines that increase insulin release. • Lower limb amputation risk. Contact a doctor about infections or ulcers on the legs or feet. • Serious allergic reactions, including anaphylaxis, swelling beneath the skin and severe peeling skin reactions. Stop Glyxambi and seek immediate medical attention if a serious allergic reaction occurs. • Severe, disabling joint pain. • Bullous pemphigoid, a blistering skin condition that may require hospital treatment. Report blisters or open skin sores to a doctor. • Heart failure is a warning for Glyxambi because it has been observed with two other medicines in the same class as linagliptin. Patients with known risk factors need assessment and monitoring.

When to call your doctor

- Seek immediate medical attention for pain, tenderness, redness or swelling around the genitals or between the genitals and anus, together with fever or feeling unwell. - Get urgent medical help for a serious allergic reaction, including swelling beneath the skin or peeling skin. - Report new skin blisters or open sores to your doctor promptly. - Contact your doctor promptly for severe or disabling joint pain. - Contact your doctor promptly for signs of infection or ulcers on your legs or feet.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.

Before taking this medicine

Do not take Glyxambi if you are allergic to empagliflozin, linagliptin or any other ingredient in the medicine. Serious allergic reactions have occurred, including anaphylaxis, swelling beneath the skin and severe peeling skin reactions. Glyxambi requires caution in people at risk of ketoacidosis, including those with type 1 diabetes. Ketoacidosis must be considered regardless of blood glucose levels. Stop Glyxambi if ketoacidosis or inflammation of the pancreas (pancreatitis) is suspected and seek urgent medical assessment. Kidney problems, older age and use of loop diuretics increase concerns about fluid loss and kidney function. Fluid levels and kidney function should be checked before treatment in these patients and monitored during treatment. Taking Glyxambi with insulin or medicines that stimulate insulin release can increase the risk of low blood sugar. Other warnings include serious urinary and genital infections, lower-limb infections or ulcers, severe joint pain, and a blistering skin condition called bullous pemphigoid. Report blisters or skin sores. Seek immediate assessment for pain, tenderness, redness or swelling around the genitals or the area between the genitals and anus, especially with fever or feeling unwell. People with risk factors for heart failure need careful assessment and monitoring. Heart failure has been observed with two other medicines in the same class as linagliptin. Glyxambi may pose a risk to an unborn baby, particularly during the second and third trimesters, and is not recommended during breastfeeding. Its safety and effectiveness in children have not been established.

Interactions

Tell your doctor if you take any of the following: - Diuretics (water pills): taking them with Glyxambi can increase urination and the risk of dehydration. Your doctor should check your fluid levels and kidney function before treatment and monitor them during treatment. - Insulin or medicines that stimulate insulin release, such as sulfonylureas: the combination increases the risk of low blood sugar. Your prescriber may need to adjust these medicines. - Lithium: Glyxambi may lower lithium levels in your blood. More frequent blood tests are needed when Glyxambi is started or its dose is changed. - Rifampin or other medicines that strongly increase the activity of P-glycoprotein or CYP3A4: these may make linagliptin less effective. Alternative treatments are strongly recommended with these medicines. Glyxambi causes glucose to pass into the urine, leading to positive urine glucose tests. Urine glucose tests and the 1,5-anhydroglucitol (1,5-AG) blood test should not be used to monitor blood sugar control during treatment; other monitoring methods are needed.

Dosing

Storage

Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
