About Gleevec
Overview
Gleevec contains imatinib, a prescription kinase inhibitor supplied as tablets. It is used to treat certain forms of Philadelphia chromosome-positive leukemia, specific blood disorders, and tumors including Kit (CD117)-positive gastrointestinal stromal tumors (GIST) and dermatofibrosarcoma protuberans (DFSP). Its use depends on the condition, disease stage and, in some cases, specific genetic findings. Imatinib works by blocking certain proteins called tyrosine kinases that help affected cells grow and multiply.
What it treats
• Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML): newly diagnosed chronic-phase disease in adults and children; blast crisis or accelerated-phase disease; or chronic-phase disease after interferon-alpha treatment has failed. • Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL): disease that has returned or resisted treatment in adults, or newly diagnosed disease in children, alongside chemotherapy. • Myelodysplastic/myeloproliferative diseases (MDS/MPD) with PDGFR gene rearrangements in adults. • Aggressive systemic mastocytosis in adults without the D816V c-Kit mutation, or when c-Kit mutation status is unknown. • Hypereosinophilic syndrome and/or chronic eosinophilic leukemia in adults, whether FIP1L1-PDGFRα fusion kinase status is positive, negative or unknown. • Dermatofibrosarcoma protuberans in adults when it cannot be removed surgically, has returned or has spread. • Kit (CD117)-positive malignant gastrointestinal stromal tumors (GIST) that cannot be removed surgically or have spread. • Kit (CD117)-positive GIST in adults as additional treatment after surgical removal.
Safety information
Side effects
Common side effects include swelling from fluid retention, nausea, vomiting, muscle cramps, muscle or joint pain, diarrhea, rash, tiredness and abdominal pain. Each was reported in at least 30% of patients. Contact a doctor promptly about unexpected rapid weight gain or other serious side effects. These include severe fluid retention; low red blood cell, white blood cell or platelet counts; heart failure or reduced heart pumping function; severe liver damage; serious bleeding; a hole in the stomach or intestine; and severe blistering skin reactions, including Stevens-Johnson syndrome. Some cases of liver damage and gastrointestinal perforation have been fatal. Other serious risks include reduced kidney function, tumor lysis syndrome (complications from the rapid breakdown of tumor cells), and severe heart problems when treatment begins in people with high eosinophil levels. An underactive thyroid has been reported in people who have had their thyroid removed and take levothyroxine. Slowed growth has been reported in children and pre-adolescents. Treatment requires monitoring of blood counts, liver and kidney function, weight and, in children, growth. Imatinib can harm an unborn baby; effective contraception is advised. Use caution when driving or operating machinery, as impairments and motor vehicle accidents have been reported during treatment.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Interactions
Some medicines can lower or raise imatinib levels in the blood by affecting an enzyme called CYP3A4. Imatinib can also raise blood levels of other medicines processed by CYP3A4 or CYP2D6. Ask your doctor to check your other medicines for these interactions. Tell your doctor if you take warfarin or need an anticoagulant. The label recommends low-molecular-weight or standard heparin rather than warfarin. Imatinib can slow the removal of high-dose methotrexate from the body. Tell your doctor if you receive methotrexate.

Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
