About Bavencio
Overview
Bavencio (avelumab) is a prescription antibody medicine used to treat Merkel cell carcinoma that has spread to other parts of the body in adults and children aged 12 years and older. It also treats locally advanced or metastatic urothelial carcinoma, either as maintenance treatment when cancer has not progressed after initial platinum-containing chemotherapy, or when cancer has progressed during or after platinum-containing chemotherapy, including within 12 months of such treatment given before or after surgery. It is used with axitinib as the first treatment for advanced renal cell carcinoma, a type of kidney cancer. Bavencio blocks a protein called PD-L1, helping restore the immune system’s ability to respond to cancer cells.
What it treats
- Metastatic Merkel cell carcinoma in adults and children aged 12 years and older. - Locally advanced or metastatic urothelial carcinoma: as maintenance treatment when the cancer has not progressed with first-line platinum-containing chemotherapy, or when it has progressed during or after platinum-containing chemotherapy, including within 12 months of such treatment given before or after surgery. - Advanced renal cell carcinoma, as first-line treatment in combination with axitinib.
Safety information
Side effects
Common side effects vary with the cancer being treated and whether Bavencio is used with axitinib. The label reports the following in at least 20% of patients in the relevant treatment groups: • Merkel cell carcinoma: tiredness, muscle and bone pain, infusion-related reactions, rash, nausea, constipation, cough and diarrhea. • Urothelial cancer during maintenance treatment: tiredness, muscle and bone pain, urinary tract infection and rash. • Previously treated urothelial cancer: tiredness, infusion-related reactions, muscle and bone pain, nausea, decreased appetite and urinary tract infection. • Renal cell carcinoma when used with axitinib: diarrhea, tiredness, high blood pressure, muscle and bone pain, nausea, inflammation of the mouth or other mucous membranes, redness and soreness of the palms and soles, voice changes, decreased appetite, an underactive thyroid, rash, liver toxicity, cough, shortness of breath, abdominal pain and headache. Serious side effects can be severe or fatal. Contact a doctor promptly if you experience or suspect a serious reaction. Bavencio can cause the immune system to attack healthy organs or tissues, including the lungs, bowel, liver, hormone-producing glands, kidneys and skin. It can also cause rejection of a transplanted organ. Blood tests to check liver, kidney and thyroid function are needed before and during treatment. Infusion-related reactions may require the infusion to be interrupted, slowed or stopped permanently. Serious, sometimes fatal complications can occur in people who receive a donor stem cell transplant before or after treatment. Major heart and blood vessel events are also a risk when Bavencio is used with axitinib. Bavencio can harm an unborn baby. Use effective contraception if you can become pregnant, and contact your doctor if you become pregnant during treatment.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
The FDA label lists no contraindications for Bavencio. However, important cautions apply: Bavencio can cause severe or fatal immune-related reactions in any organ or tissue, including the lungs, bowel, liver, hormone-producing glands, kidneys and skin. It may also cause rejection of a transplanted organ. Liver enzymes, kidney function and thyroid function should be checked before and periodically during treatment. Treatment may need to be withheld or permanently stopped depending on the reaction. Infusion-related reactions may require the infusion to be slowed, interrupted or permanently stopped. Serious or fatal complications can occur in people who receive an allogeneic hematopoietic stem cell transplant before or after treatment with a PD-1/PD-L1 blocking antibody. Major cardiovascular events are a concern when Bavencio is used with axitinib. Cardiovascular risk factors should be managed, and the combination must be stopped for severe events. Bavencio can harm an unborn baby. People who could become pregnant should use effective contraception. Do not breastfeed during treatment.
