About Balversa
Overview
Balversa contains erdafitinib, a kinase inhibitor supplied as tablets. It treats adults with locally advanced or metastatic urothelial cancer that has certain FGFR3 gene changes and has progressed during or after at least one previous systemic treatment; an FDA-approved test is used to identify eligible patients. It is not recommended for patients who are eligible for PD-1 or PD-L1 inhibitor therapy but have not yet received it. It works by blocking FGFR proteins involved in cell signaling.
What it treats
- Locally advanced or metastatic urothelial carcinoma in adults with susceptible FGFR3 genetic alterations, after the cancer has progressed during or after at least one prior systemic treatment. Eligibility is determined using an FDA-approved companion diagnostic test. Balversa is not recommended for patients who are eligible for PD-1 or PD-L1 inhibitor therapy but have not yet received it.
Safety information
Side effects
Common side effects include nail problems, mouth sores, diarrhea, tiredness, dry mouth, dry skin, reduced appetite, changes in taste, constipation, dry eyes, hair loss, and redness or soreness of the palms and soles. Common blood test changes include increased or decreased phosphate, increased calcium or potassium, decreased sodium or hemoglobin, and increased creatinine or liver enzymes. Balversa can cause serious eye problems, including fluid buildup under the retina or separation of a layer beneath it. Contact your doctor if you notice any vision changes. Eye examinations are needed monthly for the first four months of treatment, then every three months, and whenever visual symptoms occur. High phosphate levels also require monitoring and may require your prescriber to adjust treatment. Balversa can harm an unborn baby. Use effective contraception and contact your doctor if you become pregnant during treatment.

Before taking this medicine
The label lists no contraindications for Balversa. However, important precautions apply: Balversa can cause eye problems involving the retina. Eye examinations are required monthly for the first four months, then every three months, and whenever visual symptoms occur. Treatment may need to be paused or permanently stopped if these problems develop. Balversa can increase phosphate levels in the blood. These levels need monitoring, and the prescriber may need to adjust treatment. Balversa can harm an unborn baby. Patients should be advised of this risk and use effective contraception. Do not breastfeed during treatment.
Interactions
Tell your doctor about all medicines and supplements you take, particularly: • Medicines that moderately block CYP2C9 or strongly block CYP3A4, enzymes involved in processing medicines. Your doctor may choose alternatives or monitor you closely for side effects. • Strong CYP3A4 inducers, which increase this enzyme’s activity. These should not be taken with Balversa. Moderate CYP3A4 inducers also require your prescriber to review treatment. • Products that change blood phosphate levels. These should be avoided before the initial dose-adjustment period. • Medicines called P-glycoprotein (P-gp) substrates that have a narrow safety margin. These need to be taken separately from Balversa; ask your doctor about the required timing.

Dosing
Dosing is set by your prescriber. Each health care professional may prescribe medication differently according to a patient's medical history and condition, so follow the directions on your prescription label and speak to your doctor or pharmacist before changing anything.
Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
