About Abraxane
Overview
Abraxane is a prescription cancer medicine containing paclitaxel bound to human albumin, supplied in a vial for preparation and infusion into a vein. It treats breast cancer that has spread after combination chemotherapy has failed or the cancer has returned within six months of adjuvant chemotherapy; prior treatment should have included an anthracycline unless medically unsuitable. It is also used as first-line treatment with carboplatin for locally advanced or metastatic non-small cell lung cancer when curative surgery or radiation is not an option, and with gemcitabine for metastatic pancreatic adenocarcinoma. It works by interfering with the normal rearrangement of microtubules, structures inside cells needed for cell division and other vital functions.
What it treats
• Metastatic breast cancer after combination chemotherapy has failed, or when the cancer returns within 6 months of adjuvant chemotherapy. Previous treatment should have included an anthracycline unless medically unsuitable. • Locally advanced or metastatic non-small cell lung cancer, as first-line treatment with carboplatin, when surgery or radiation therapy aimed at a cure is not suitable. • Metastatic adenocarcinoma of the pancreas, as first-line treatment with gemcitabine.
Safety information
Side effects
Common side effects vary depending on the cancer being treated and whether Abraxane is used alone or with other medicines. They include hair loss, tiredness or weakness, nausea, diarrhea, low white blood cell counts, anemia (low red blood cell counts), and nerve problems affecting sensation. Other common effects reported with Abraxane alone for metastatic breast cancer include muscle or joint pain, infections, abnormal electrical heart recordings (ECGs), and raised liver enzyme levels. Low platelet counts are common when used with carboplatin for non-small cell lung cancer. With gemcitabine for pancreatic cancer, common effects also include swelling, fever, vomiting, reduced appetite, rash, and dehydration. Contact your doctor promptly about fever, infections, nerve problems, dehydration, or vomiting. Nerve problems may require treatment changes or interruption. Serious reactions include severe reductions in blood cell counts and pneumonia. Sepsis and pneumonitis (lung inflammation) have occurred with Abraxane plus gemcitabine; sepsis can occur even without a low white blood cell count. Severe allergic reactions, including fatal reactions, have been reported. Seek urgent medical help for a severe allergic reaction. Abraxane must not be given again after such a reaction. Liver impairment can increase exposure to paclitaxel and the risk of toxicity, so close monitoring is needed.

Before taking this medicine
Do not use Abraxane if your neutrophil count (a type of white blood cell) is below 1,500 cells/mm³ before treatment, or if you have previously had a severe allergic reaction to Abraxane. Severe allergic reactions, including fatal reactions, have been reported. Liver impairment requires caution and close monitoring because it can increase exposure to paclitaxel and its toxic effects. Sensory nerve damage occurs frequently and may require treatment changes or interruption. When used with gemcitabine, Abraxane has been associated with sepsis, with or without low neutrophil counts, and pneumonitis (lung inflammation). Sepsis requires treatment interruption; pneumonitis requires permanent discontinuation of both medicines. Abraxane can harm an unborn baby. Use effective contraception during treatment and do not breastfeed. Abraxane contains albumin derived from human blood, which carries a theoretical risk of viral transmission.
Interactions
Abraxane requires caution when used with medicines that block or increase the activity of CYP2C8 or CYP3A4, enzymes involved in breaking down paclitaxel. Ask your doctor to review your other medicines for these interactions.
