About Gleolan
Overview
Gleolan (aminolevulinic acid hydrochloride) is an optical imaging agent that helps surgeons see malignant tissue during brain surgery. It is used in patients with glioma suspected to be World Health Organization grade III or IV on imaging before surgery. It causes a substance called PpIX to build up in tumor cells, making the tissue glow red under a specially adapted surgical microscope with blue light.

Used to help surgeons see malignant tissue during surgery in patients with glioma suspected to be WHO grade III or IV on preoperative imaging.
Safety information
Side effects
Side effects reported in more than 1% of patients during the week after surgery were fever, low blood pressure, nausea and vomiting. Less frequently reported effects during the first six weeks after surgery included chills, sensitivity to light, skin inflammation from sunlight, abnormal liver function tests and diarrhoea. Raised liver enzymes occurred in clinical studies; no cases of liver failure were reported. Seek medical attention promptly for difficulty speaking, weakness or paralysis, loss of part of your vision, reduced sensation or seizures. These were among the neurological events related to the surgical procedure, which occurred in 29% of patients. Headache and brain swelling were also reported among these events. Gleolan can cause reactions to light. Reduce exposure to sunlight and room lights for 48 hours after receiving it. Contact your doctor if you develop a skin reaction to light.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
Gleolan must not be used in people who are allergic to aminolevulinic acid (ALA) or porphyrins, or who have acute or chronic porphyria. Medicines that can cause reactions to light must not be given during the 24-hour period around surgery. These include St. John’s wort, griseofulvin, thiazide diuretics, sulfonylureas, phenothiazines, sulphonamides, quinolones and tetracyclines. Topical preparations containing ALA must also be avoided during this period. Exposure to sunlight and room lights should be reduced for 48 hours after taking Gleolan. During surgery, tissue that does not fluoresce may still contain tumour.
Interactions
Medicines and supplements that increase sensitivity to light can raise the risk of a phototoxic skin reaction (severe sunburn) with Gleolan. Tell your doctor if you use St. John’s wort, griseofulvin, thiazide diuretics, sulfonylureas, phenothiazines, sulfonamides, quinolones, tetracyclines, or skin preparations containing aminolevulinic acid (ALA). These should be avoided for 24 hours before and after Gleolan administration; discuss any necessary changes with your doctor.

Storage
Store in the original carton below 25°C, protected from light, unless the pack states refrigeration. Do not freeze. Keep all medicines out of the sight and reach of children.
