Increased risk of serious bacterial, fungal, viral and opportunistic infections leading to hospitalization or death, including tuberculosis (TB). Discontinue treatment with CIBINQO if serious or opportunistic infection occurs. Test for latent TB before and during therapy; treat latent TB prior to use. Monitor all patients for active TB during treatment, even patients with initial negative latent TB test.
Higher rate of all-cause mortality, including sudden cardiovascular death, with another JAK inhibitor vs. TNF blockers in rheumatoid arthritis (RA) patients. CIBINQO is not approved for use in RA patients.

About Cibinqo
Overview
Cibinqo (abrocitinib) is a Janus kinase (JAK) inhibitor supplied as tablets. It treats moderate-to-severe atopic dermatitis in adults and children aged 12 years and older when other systemic medicines, including biologics, have not controlled the condition adequately or are not advisable. It works by blocking the activity of an enzyme called JAK1, although how inhibition of specific JAK enzymes contributes to its treatment effect is not fully understood. It is not recommended for use with other JAK inhibitors, biologic immune-modifying medicines or other immunosuppressants.
What it treats
Moderate-to-severe atopic dermatitis in adults and children aged 12 years and older that is not adequately controlled by other systemic medicines, including biologics, or when those treatments are not advisable.
Safety information
Side effects
Common side effects include cold-like symptoms, nausea, headache, herpes simplex infections, dizziness, urinary tract infections, tiredness, acne, vomiting, impetigo (a skin infection), throat pain, high blood pressure, flu, stomach or bowel infections, upper stomach pain, stomach discomfort, shingles and contact dermatitis. Blood tests may show increased creatine kinase levels or a low platelet count. Serious risks include serious infections, cancer and abnormal growth of lymphatic cells, major heart or blood vessel events, and blood clots. These require prompt medical attention. The label also warns about a risk of death. Cibinqo can cause changes in platelet counts, lymphocyte counts and blood fats, so laboratory monitoring is recommended. Low blood sugar can occur in people with diabetes; closer blood sugar monitoring may be needed. If you have diabetes, contact your healthcare provider if you develop signs or symptoms of low blood sugar.

Before taking this medicine
Do not take Cibinqo with antiplatelet medicines during the first 3 months of treatment, except for low-dose aspirin. Cibinqo is not recommended for people with severe kidney impairment, end-stage kidney disease or severe liver impairment. Breastfeeding is not recommended during treatment. If you have diabetes, more frequent blood glucose monitoring may be needed. Tell your healthcare provider if you develop signs or symptoms of low blood sugar. Blood tests are recommended to monitor for changes in platelets, lymphocytes and blood lipid levels. Avoid live vaccines immediately before, during and immediately after treatment.
Interactions
Tell your doctor about all medicines you take before using Cibinqo. Strong inhibitors of the enzyme CYP2C19 may require your prescriber to adjust your Cibinqo dose. Avoid taking Cibinqo with medicines that moderately or strongly inhibit both CYP2C19 and CYP2C9, or that moderately or strongly increase the activity of either enzyme. Some medicines that are P-gp or CYP2C19 substrates may need monitoring or dose adjustment because small changes in their levels can cause serious or life-threatening toxicity. Ask your doctor whether any of your medicines fall into these groups.

Dosing
Dosing is set by your prescriber. Each health care professional may prescribe medication differently according to a patient's medical history and condition, so follow the directions on your prescription label and speak to your doctor or pharmacist before changing anything.
Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
