FDA boxed warningRisk of heart failure
CAMZYOS reduces left ventricular ejection fraction (LVEF) and can cause heart failure due to systolic dysfunction.
Echocardiogram assessments of LVEF are required prior to and during treatment with CAMZYOS. Initiation of CAMZYOS in patients with LVEF <55% is not recommended. Interrupt CAMZYOS if LVEF is <50% at any visit or if the patient experiences heart failure symptoms or worsening clinical status.
Concomitant use of CAMZYOS with certain cytochrome P450 inhibitors or discontinuation of certain cytochrome P450 inducers may increase the risk of heart failure due to systolic dysfunction; therefore, the use of CAMZYOS is contraindicated with the following:
Strong CYP2C19 inhibitors
Moderate to strong CYP2C19 inducers or moderate to strong CYP3A4 inducers
Because of the risk of heart failure due to systolic dysfunction, CAMZYOS is available only through a restricted program under a Risk Evaluation and Mitigation Strategy (REMS) called CAMZYOS REMS PROGRAM.
CAMZYOS can cause heart failure due to systolic dysfunction.

About Camzyos
Overview
Camzyos contains mavacamten, a cardiac myosin inhibitor. It is used to improve symptoms and the ability to be physically active in adults with symptomatic obstructive hypertrophic cardiomyopathy (HCM), classified as NYHA class II–III. It works by reducing excessive interaction between proteins that make the heart muscle contract, helping reduce obstruction to blood leaving the heart and improving heart filling pressures.
What it treats
Treatment of symptomatic obstructive hypertrophic cardiomyopathy (HCM) in adults with New York Heart Association (NYHA) class II–III symptoms, to improve functional capacity and symptoms.
Safety information
Side effects
Common side effects include dizziness and fainting. Camzyos can cause heart failure, a serious risk that requires medical attention. Contact your doctor if you faint or suspect heart failure. Interactions with other medicines, including over-the-counter products, can increase the risk of heart failure or reduce Camzyos’s effectiveness. Camzyos may also harm an unborn baby. Contact your doctor if you become pregnant during treatment.

Before taking this medicine
Do not take Camzyos with medicines that strongly inhibit CYP2C19, or with medicines that moderately or strongly induce CYP2C19 or CYP3A4. Ask your prescriber to check all medicines you use, including over-the-counter products, because interactions may lead to heart failure or make Camzyos less effective. Camzyos carries a risk of heart failure. If you develop another illness during treatment, your prescriber may consider temporarily interrupting treatment. Camzyos may harm an unborn baby. If you could become pregnant, use effective contraception during treatment and for 4 months after the last dose. Avoid combined hormonal contraceptives containing a progestin other than norethindrone.
Interactions
Tell your doctor about all medicines you take. Medicines that weakly or moderately inhibit CYP2C19, or moderately or strongly inhibit CYP3A4, may increase the risk of heart failure with Camzyos. Starting one of these medicines requires your prescriber to reduce your Camzyos dose and arrange additional monitoring. Medicines that reduce the force of the heart’s contractions (negative inotropes) require close medical supervision and monitoring of how well the heart pumps if they are started or their dose is increased. Some combinations with Camzyos should be avoided.

Storage
