About Afinitor Disperz
Overview
Afinitor Disperz contains everolimus, a kinase inhibitor. It treats people aged 1 year and older with tuberous sclerosis complex (TSC) who have a subependymal giant cell astrocytoma (SEGA) that needs treatment but cannot be completely removed by surgery. It is also used alongside other treatment for TSC-associated partial-onset seizures in people aged 2 years and older. It works by blocking mTOR, a protein involved in cell growth and nerve cell activity that can be overactive in TSC.
What it treats
- Subependymal giant cell astrocytoma (SEGA) associated with tuberous sclerosis complex (TSC) in adults and children aged 1 year and older, when treatment is needed but surgery cannot completely remove the tumour. - Partial-onset seizures associated with TSC in adults and children aged 2 years and older, as an add-on to other seizure treatment.
Safety information
Side effects
Common side effects vary by the condition being treated. They include mouth sores and inflammation, respiratory tract infections and other infections, rash, tiredness or weakness, diarrhoea, swelling, abdominal pain, nausea, fever, cough, headache and reduced appetite. Serious side effects include lung inflammation not caused by infection, severe allergic reactions, angioedema (swelling beneath the skin), kidney failure, impaired wound healing, increased blood sugar or blood fats, reduced blood cell production, and severe reactions related to radiation treatment. Infections can also be serious. Taking an ACE inhibitor alongside this medicine may increase the risk of angioedema. Contact your doctor promptly if you develop signs of infection, breathing problems, an allergic reaction, swelling or difficulty with wound healing. Blood tests are needed before and during treatment to monitor kidney function, blood sugar, blood fats and blood cell counts, as some problems may not cause noticeable symptoms.

Before taking this medicine
Do not take Afinitor Disperz if you have a clinically significant allergy to everolimus or other rapamycin derivatives. Conditions requiring caution include lung inflammation not caused by infection, infections, kidney or liver problems, high blood sugar or lipid levels, and low blood cell counts. Kidney function, blood sugar, lipids and blood counts should be checked before and during treatment. Older patients need monitoring for side effects. Taking an ACE inhibitor alongside this medicine may increase the risk of angioedema, a serious swelling reaction. Clinically significant allergic reactions or angioedema require permanent discontinuation. Afinitor Disperz can impair wound healing. Treatment needs to be withheld around planned surgery, with restarting guided by adequate wound healing. Severe radiation reactions may occur. Avoid live vaccines and close contact with people who have received them. Recommended childhood vaccinations should be completed before treatment. Afinitor Disperz can harm an unborn baby. Patients who could conceive should use effective contraception.
Interactions
Tell your doctor if you take cannabidiol oral solution or medicines that affect the CYP3A enzyme and P-gp transporter. Avoid taking Afinitor Disperz with strong CYP3A and P-gp inhibitors. Moderate inhibitors, strong inducers, and cannabidiol oral solution may require your prescriber to adjust your Afinitor Disperz dose. Ask your doctor whether any of your medicines fall into these groups; do not change your dose yourself.

Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
